Abstract

Background:
HDT-BR offers a model to examine the effect of immobilization on cartilage biomarkers. The aim of this study was to observe serum concentrations of type II procollagen carboxy-propeptide (CPII) and type II collagen (CII)-related neoepitope (C2C) in response to HDT-BR and to investigate how possible changes are affected by resistive vibration exercise (RVE) alone or in combination with a protein enhanced diet (NEX).
Methods:
Twelve healthy male subjects (age 34.2 ± 8.3 years; BMI 22.4 ± 1.7 kg/m²) performed 21-days of HDT-BR with a cross-over design (Control (CON), CON+RVE, or CON+NEX) in three campaigns (C1, C2, C3). Serum samples were taken during baseline data collection (BDC) day-3 to -1, during HDT2, HDT3, HDT5, HDT7, HDT14 and HDT21, and during the recovery R1 and R6. sCPII and sC2C was measured using Enzyme-linked immunosorbent assays (ELISA, IBEX Technologies Inc., Montreal, Canada). Linear Mixed-Effects Models were used to detect significant changes with the predictors.
Results:
At BDC-3 of C1, mean concentration was 1319.3(±396.0) ng/mL for sCPII, and 95.2(±23.8) ng/mL for sC2C. Time points(p<0.001) and campaign(p<0.001) affected serum concentrations for both markers. sCPII at successive time points during HDT-BR (HDT2, HDT3, HDT5, HDT7, HDT14) was significantly higher compared to baseline, increasing by 15.9% at HDT2 (p=0.001) and 18.4% at HDT5 (p<0.001). Similar increases were noted for C2C at all HDT-BR time points, with the highest increase (+35.8%) compared to BDC-3 at HDT7(p<0.001). C2C was still elevated on R1 (p=0.001) and R6(p<0.001). When sC2C increased by one unit, sCPII increased by 6.44 ng/mL(p<0.001). After the first 72h of HDT-BR, the ratio CPII:C2C decreases continuously, with a highest decrease of 13.1% on HDT21(p=0.004). RVE and NEX significantly affected sCPII at certain levels of campaign (C2, C3) with notable changes.
Conclusion:
BR caused substantial changes of sCPII and sC2C; while baseline values were in the range of healthy individuals, serum concentrations were higher after HDT-BR.
