Ample evidence suggests beneficial effects of physical activity and exercise on cancer development and mortality. The knowledge about the underlying mechanisms is still sparse. It is hypothesized that exercise induces an activation of the immune system with an enhancement of the body´s own tumor defense. Especially natural killer cells (NK-cells) are of interest, since they can recognize and eliminate neoplastic cells. A vast amount of literature showed that NK-cells are regulated during differentiation and activation by epigenetic modifications. Furthermore, physical activity and exercise induce epigenetic changes in various tissues, leading to functional adaptions. Therefore this thesis aims to analyze the effect of a single bout of endurance exercise on DNA-methylation in isolated NK-cells.
In this thesis two studies were conducted. The first study analyses the promoter DNA-methylation and gene expression of two NK-cell-receptors (activating and inhibiting), before and after a single bout of endurance exercise. The activating receptor showed a lower DNA-methylation and a higher gene expression after the acute exercise bout. In contrast, the inhibiting receptor was not influenced by exercise. In the second study the influence of a single bout of exercise on genome-wide DNA-methylation was assessed. The acute exercise bout changed the DNA-methylation of 25 genes. Whether or not these changes were accompanied by alterations of gene expression or cell function is not known.
In summary, this thesis could show that an acute bout of endurance exercise changes the DNA-methylation of isolated NK-cells that at least partially leads to a change in gene expression. The used combination of very detailed single gene analyses and genome-wide analyses leads to a good description of the exercise effects. Moreover, these studies are the first performing such analyses in a precise cell population rather than using cell conglomerates. Therefore, these data could lead to a better understanding of the exercise-induced epigenetic changes in NK-cells.
The described studies should motivate further analyses in this research field in order to gain an understanding on the exercise-induced changes in epigenetic modification of NK-cells. In view of dose response relations, different exercise modalities should be used. Finally, these examinations need to be confirmed wit functional analyses.
